I'm fairly surprised no one's mentioned Luna, but I'm thinking people were under the impression it had to be mane 6.

Oh, I am most certainly a big fan of Luna/Nightmare Moon. She was my first fan-art:

I love Fluttershy, and I really like Rarity too, but I would have to give my vote to Pinkie Pie as the best pony because she was the pony that addicted me, and frankly, you can't do better than that.
